Analogs of oxytocin containing a modified peptide bond.

چکیده
Analogs of deamino-oxytocin and deamino-oxypressin containing a CH2-NH group instead of an amide bond between positions 8 and 9 were synthesized. All tested compounds exhibit significantly lowered biological activities.

Conformationally biased analogs of oxytocin.
Four diastereomeric analogs of oxytocin containing substituted phenylalanine in position 2 were synthesized. This modified phenylalanine side chain contained one methyl group attached to the beta-carbon and the second one at the 2' position of the aromatic ring. All analogs were found to be inhibitors of uterotonic activity of oxytocin with pA2 values ranging from 6.0 to 8.3; the most potent on...
متن کاملCollision-induced liquid secondary-ion mass spectra of oxytocin derivatives. A study of the influence of modifications in the disulfide bond on the adjacent peptide bond cleavage.
The influence of the inductive effect of modifications in the disulfide bridge in oxytocin analogs on the cleavage of the neighboring peptidic bond was studied. A correlation between the inductive constant of the group in the disulfide bridge and the particular fragment ion abundance was observed.

Analogs of oxytocin containing tetrahydroisoquinoline carboxylic acid (Tic) of L or D configuration in position 2 were synthesized and their biological activities were tested. Both analogs showed negligible agonist activity in uterotonic, galactogogic, and pressor assays, but they are in vitro uterotonic inhibitors. In comparison with oxytocin analogs containing L- or D-phenylalanine in positio...
متن کاملBiofunctional evaluation of a hydrogen bond linking the ring and tail beta-turns of oxytocin.
Deamino-[8-N-methylleucine]oxytocin and deamino-[8-alpha-hydroxyisocaproic acid]oxytocin were synthesized to study the importance of hydrogen bonding between the carboxamide carbonyl of asparagine and the peptide N-H of leucine in stabilizing the biologically active conformation of oxytocin. The analogs were synthesized by coupling deaminotocinoic acid with Pro-Leu(Me)-Gly-NH2 and Pro-HyIc-Gly-...
متن کاملProton n.m.r. spectroscopic evidence for sulfur-aromatic interactions in peptides.
The downfield shift of the tyrosyl proton resonances and an increased chemical shift difference between the resonances for the 2',6' and 3',5' hydrogens in a series of deamino-oxytocin analogs modified in the disulfide bridge provide evidence for aromatic-sulfur interactions in d6-dimethylsulfoxide solutions.
